The sample was pre-dried at 105°C to constant weight, 0.4 g of the dried sample was weighed, 50 mL of glacial acetic acid was added, and heated to dissolve. Add 25mL mercuric acetate solution, cool, add 2 drops of crystal violet indicator, titrate with perchloric acid standard solution (0.1mol/L) until the solution turns green, and correct the titration result with blank test.
Colorimetric method: Under the condition of pH=1.0, betaine hydrochloride can form a red precipitate with Ray's salt. After centrifugation, after discarding the supernatant, the precipitate is dissolved in 70% acetone and becomes a pink solution. The maximum absorption peak appears at 525 nm. When the content of betaine hydrochloride is 0.1~12.5mg, it conforms to the Lambert-Beer law.
High performance liquid chromatography: suitable for the determination of betaine content in wolfberry. The betaine component in the sample was extracted with methanol solution (2.5+7.5), purified with mixed cation exchange solid phase extraction column, and then fixed with acetonitrile solution (7.5+2.5), and finally with a UV detector. High-performance liquid chromatograph was used for determination at a wavelength of 205 nm, qualitative determination was performed according to the retention time of chromatographic peaks, and external standard method was used for quantitative determination.
